Secondary Antibodies
The reactivity of the antiserum is directed to the Fc subunit of the IgG molecule including all subclasses. In immunoelectrophoresis and double radial immunodiffusion, using various antiserum concentrations against normal rat plasma and serum, a characteristic precipitin line is obtained which shows a reaction of identity with the precipitin line obtained with purified IgG. No reaction is obtained with purified IgA, IgM and IgG/Fab fragments or any other serum protein. In immunoelectrophoresis and double radial immunodiffusion to identify the presence of IgG; as secondary antibody to precipitate the immunoglobulin in normal rat serum; to prepare an immunoadsorbent for the production of Ig-free rat serum, plasma or non-Ig preparations.
Purified normal IgG, including all known subclasses, isolated from pooled rat serum. Freund’s complete adjuvant is used in the first step of the immunization procedure.
Delipidated, heat inactivated lyophilized stable whole serum No preservative added, as it may interfere with the antibody activity. Total protein and IgG concentration in the antiserum are comparable to those of pooled goat serum. No foreign proteins added.

Inter-species cross-reactivity is a normal feature of antibodies to immunoglobulins, since Ig of different species frequently share antigenic determinants. Cross-reactivity of this antiSerum has not been tested in detail, however in double radial immunodiffusion a reaction with Mouse has been obtained.

Immunoprecipitation
Immunoprecipitation. In immunoelectrophoresis use 2 μl or equivalent against 120 μl antiserum. In double radial immunodiffusion (Ouchterlony) use a rosette arrangement with 10 μl antiserum in a 3 mm diameter centre well and 2 μl serum samples (neat and diluted) in 2 mm diameter peripheral wells. Precipitin titre not less than 1:16 when tested against normal rat serum in agar block titration.
The lyophilized antiserum is shipped at ambient temperature and may be stored at +4°C; prolonged storage at or below -20°C. It is reconstituted by adding 1 ml sterile distilled water, spun down to remove insoluble particles, divided into small aliquots, frozen and stored at or below -20°C. Prior to use, an aliquot is thawed slowly at ambient temperature, spun down again and used to prepare working dilutions by adding sterile phosphate buffered saline (PBS, pH 7.2). Repeated thawing and freezing should be avoided. Working dilutions should be stored at +4°C, not refrozen, and preferably used the same day. If a slight precipitation occurs upon storage, this should be removed by centrifugation. It will not affect the performance of the product. Lyophilized at +4° C--at least 10 years. Reconstituted at or below -20° C--3-5 years. Reconstituted at +4° C--7 days

This antibody needs to be stored at + 4°C in a fridge short term in a concentrated dilution. Freeze thaw will destroy a percentage in every cycle and should be avoided.
