Long description

The reactivity of the antiserum is directed to the Fc subunit of the IgE molecule, which expresses strict (class) specificity. In immunoelectrophoresis and radial in immunodiffusion using various antiserum concentrations against human serum, a single precipitin line has been obtained which shows a reaction of identity with the precipitin lines obtained with the purified IgE used as immunogens. It does not react with IgG including all subclasses, IgG/Fab fragments, IgM and IgA or any non-Ig protein in human serum, as tested by immunoelectrophoresis and double radial immunodiffusion. The antiserum also react with membrane-bound IgE in peripheral blood cells as tested by immunofluorescence microscopy. In immunocytochemical and immunohistochemical staining for the detection of IgE at the cellular and subcellular level by staining of appropriately treated cell and tissue substrates; to identify and measure IgE in human serum or other body fluids. This immunoconjugate is not pre-diluted. The optimum working dilution of each conjugate should be established by titration before being used. Excess labelled antibody must be avoided because it may cause high unspecific background staining and interfere with the specific signal. Working dilutions are usually between 1:20 and 1:80, depending on the method used.

Antibody come from

Pools of purified homogenous IgE isolated from human serum. Freund’s complete adjuvant is used in the first step of the immunization procedure.

Other description

FITC-coupled purified hyperimmune goat IgG lyophilized from a solution in phosphate buffered saline (PBS, pH 7.2). No preservative added, as it may interfere with the antibody activity.

This immunoconjugate is not species-specific since inter-species cross-reactivity is a normal feature of antisera to immunoglobulins. Cross-reactivity of this conjugate has not been tested in detail.

Storage

The lyophilized conjugate is shipped at ambient temperature and may be stored at +4°C; prolonged storage at or below -20°C. It is reconstituted by adding 1 ml sterile distilled water, spun down to remove insoluble particles, divided into small aliquots, frozen and stored at or below -20°C. Prior to use, an aliquot is thawed slowly at ambient temperature, spun down again and used to prepare working dilutions by adding sterile phosphate buffered saline (PBS, pH 7.2). Repeated thawing and freezing should be avoided. Working dilutions should be stored at +4°C, not refrozen, and preferably used the same day. If a slight precipitation occurs upon storage, this should be removed by centrifugation. It will not affect the performance of the immunoconjugate. Lyophilized at +4°C--at least 10 years. Reconstituted at or below -20°C--3-5 years. Reconstituted at +4°C--7 days.

Warnings

This product is intended FOR RESEARCH USE ONLY, and FOR TESTS IN VITRO, not for use in diagnostic or therapeutic procedures involving humans or animals. This product contains sodium azide. To prevent formation of toxic vapors, do not mix with strong acidic solutions. To prevent formation of potentially explosive metallic azides in metal plumbing, always wash into drain with copious quantities of water. This datasheet is as accurate as reasonably achievable, but Nordic-MUbio accepts no liability for any inaccuracies or omissions in this information.

Description

This antibody needs to be stored at + 4°C in a fridge short term in a concentrated dilution. Freeze thaw will destroy a percentage in every cycle and should be avoided.Antibody against the Hu protein or peptide or recombinant supplied in 1 volumes. Ask for quote if you need bulk.

Properties

If you buy Antibodies supplied by nordc they should be stored frozen at - 24°C for long term storage and for short term at + 5°C.This nordc Fluorescein isothiocyanate (FITC) antibody is currently after some BD antibodies the most commonly used fluorescent dye for FACS. When excited at 488 nanometers, FITC has a green emission that's usually collected at 530 nanometers, the FL1 detector of a FACSCalibur or FACScan. FITC has a high quantum yield (efficiency of energy transfer from absorption to emission fluorescence) and approximately half of the absorbed photons are emitted as fluorescent light. For fluorescent microscopy applications, the 1 FITC is seldom used as it photo bleaches rather quickly though in flow cytometry applications, its photo bleaching effects are not observed due to a very brief interaction at the laser intercept. nordc FITC is highly sensitive to pH extremes.Human proteins, cDNA and human recombinants are used in human reactive ELISA kits and to produce anti-human mono and polyclonal antibodies. Modern humans (Homo sapiens, primarily ssp. Homo sapiens sapiens). Depending on the epitopes used human ELISA kits can be cross reactive to many other species. Mainly analyzed are human serum, plasma, urine, saliva, human cell culture supernatants and biological samples.Immunoglobulin E (IgE) kappa or Fc specific antibody is a kind of antibody (or immunoglobulin (Ig) "isotype") that has only been found in mammals. IgE is synthesized by plasma cells. Monomers of IgE consist of two heavy chains (ε chain) and two light chains, with the ε chain containing 4 Ig-like constant domains (Cε1-Cε4). IgEs play a role in allergy and response to parasite infection. High levels of IgEs caused by parasites can lower the allergic reaction of patients to allergens common present in the human environment. As such the parasite history of an allergic patient needs to be taken in consideration as a positive factor.
